Periodic Acid-Schiff (PAS) Stain

Periodic-Acid Schiff (PAS) Stain:

• Pathology stain based on the periodic acid (H+IO4-) oxidation (cis-diols to aldehydes) & Schiff-base reactions.
• Will stain anything with a cis-dihydroxy group a purple-magenta colour.
• Typically stains insoluble sugars.
• PAS staining is mainly used for highlighting structures containing a high proportion of carbohydrate macromolecules.

Tissues & Structures Stained by PAS:

• Alpha-1 protease inhibitor (antitrypsin)
• Amyloids (weakly)
• Basement membranes
• Cartilage
• Epithelial mucin (except some acid mucins)
• Fungal walls
• Glycogen
• Reticulin
• Type I collagen (weakly)
